载体使用Stbl3感受态细胞扩增质粒,可以减少载体基因重组几率。pLP/VSVG, pLP1, pLP2载体是Invitrogen公司出品的配套使用的慢病毒包装载体。第三代慢病毒包装系统组成成份有pLP1、pLP2和pLP/VSVG(或者pCMV-VSVG)。在使用的时候,按照pLP1(3):pLP2(2):pLP/VSVG(3)的比例配成1 ug/ul的第三代慢病毒包装系统package Mix。转染时,对于60mm的细胞培养皿来说,按照慢病毒基因质粒(3ug)和package mix(6 ug,也就是6 ul)的比例进行慢病毒质粒包装实验。
